Bayesian trial designs have recently seen widespread applications in early-phase clinical trials in cancer research, largely owing to the paradigm shift in drug development spurred by the success stories in immunotherapy. Many novel statistical designs have been proposed. Some are of methodological interest while some have become favorites among practitioners. In this talk, we review recent development in Bayesian designs in early phase clinical trial designs, with reflections on our own experience in an academic cancer center. We also discuss opportunities and challenges applying statistical trial designs to meet various demands in clinical trial development.
