Abstract:

The mechanisms for protection behind Remote Ischemic Perconditioning (RIPerC) are not fully understood yet. In this study, we have investigated the neuroprotective efficacy and the role of AMPK- eNOS pathways in limb RIPerC using a distal Middle Cerebral Artery occlusion (MCAo) rat model. Adult male rats with and without MCAo followed by perconditioning were included. Laser Speckle Contrast Imaging (LSCI), Western Blotting (WB), RT-PCR techniques were used to measure collateral flow augmentation, pAMPK, and eNOS expression respectively. Mann Whitney U-test was used to compare the pAMPK and eNOS between control and RIPerC treated rats. LSCI revealed that RIPerC Augmented collateral flow into distal segments compared to control rats. Our results also showed that pAMPK increased significantly (p< 0.01) in RIPerC treated rats (89.99±2.05) compared to controls (34.24±1.05). eNOS was also significantly (p< 0.01) increased in RIPerC treated rats (2.04±0.13) compared to controls (0.87±0.08). Our results suggest that RIPerC Augmented collateral flow into distal segments could be neuroprotective and may be potentially due to the activation of pAMPK- eNOS pathways.
