Title: Detecting Genetic Association in Case-Control Studies Using Backward Genotype Transmission Association Algorithm

Abstract:

During the past two decades, a lot of new technologies have been developed in molecular biology, which enable people to focus their studies based on the whole-genome scans. For example, the identification of large number of marker loci helps people precisely localize the genes that are disease susceptible. However, since the complex traits involve multiple genes with possible interactions, statistical data analysis must be designed carefully to dig out information. Recently, a novel method named Backward Haplotype Transmission Association algorithm was proposed by Lo and Zheng (2002), which used haplotype information in the case-parent trio data to detect those responsible markers and the interactions. Here we extend this method to case-control studies. Case-control study has the advantages of easy sample collection; while BHTA method has the advantages like powerfully detecting the multifactorial traits. We apply the idea of BHTA in case-control studies by using genotype information, so that the advantages of both sides are well combined. We also justify some theoretical points to support the rationale in this extension, and improve the algorithm by adding some controls.
