Abstract:

When applying the Cochran-Armitage trend test to decide whether a candidate-gene is associated with a disease, a score must be assigned to three genotypes. The optimal scores will depend on the underlying genetic model. When the mode of inheritance is known we explore the statistical properties of these optimal scores. On the other hand, when the inheritance model belongs to a family of genetic models, e.g., recessive, dominant or additive, we show that the MAX of the standardized test is quite power-robust over the entire family of models. If one restricts the set of possible tests to linear combinations, the simple MERT has higher minimum power than the optimal tests for the additive model. We illustrate the results by reanalyzing a melanoma study.
