SC07: Data Monitoring Committees (DMCs): Principles, Current Practices, and Paths Forward

This course will provide a comprehensive overview of Data Monitoring Committees (DMCs), covering their purpose, structure, operational role, and future across clinical trials and development programs, with a particular emphasis on confirmatory trials. The course will include lectures focused on both technical and operational aspects of DMCs, including their historical evolution, core principles such as independence and confidentiality, practical considerations for implementing DMCs (e.g., drafting DMC charters, preparing DMC reports, and structuring effective DMC meetings), and an overview of guidance from regulatory agencies. We will also address the role of DMCs in more complex and emerging trial settings, including adaptive designs, accelerated approval pathways, and program-level DMCs. In addition to lectures, the course will feature a 45-minute interactive session with a panel of experts from academia, industry, and regulatory backgrounds (statisticians and clinicians), providing participants with the opportunity to discuss real-world challenges, explore case studies, and engage directly with experienced practitioners. To conclude, we will dedicate 30 minutes to forward-looking discussion focusing on the future of DMCs, including potential roles for the use of Artificial Intelligence (AI) tools and the critical need for sponsors and investigators to actively support DMC mentorship to train the next generation of DMC members. We believe these efforts can significantly expand the pool of well-prepared DMC experts in the coming years.
