Title: Differential Abundance Analyzes of Pre- and Post-Metabolomic Data with Steroid Treatment for Bronchopulmonary Dysplasia

Abstract:

Metabolomics is a rapidly growing discipline focused on assessing the changes in small molecule metabolites in the biological systems. The disparity in the abundance of metabolites under different conditions characterizes the metabolite dynamics, interactions and response to a drug. Our differential abundance analysis utilizes the metabolites assayed using mass chromatography on urine specimens, before and after the steroid treatments in preterm infants with Bronchopulmonary Dysplasia. Wilcoxon Signed Rank test followed by multiple testing adjustments were completed to assess the changes in metabolite concentrations. Uracil, Kynurenine, Glucoheptulose and isohexonic acid were found the most significantly associated with the treatment (p< 0.005 each). Metabolite Set Enrichment Analysis was completed to identify the biological patterns that are significantly enriched in the metabolomic pathways. Urea cycle, ammonia recycling and malate-aspartate shuttle were three most significantly enriched (p < 0.05) pathways which are implicated in energy homeostasis. These preliminary results need to be studied further to understand the disease etiology and drug response in preterm infants.
