Title: A Sequential Design to Test Incremental Response Rates

Abstract:

A fixed series of therapies for a condition, usually an incremental intervention, is presumed to have an increasing response rate. Test subjects enrolled in a study are to receive the series of therapies until a response is observed. We devise a sequential study that can test incremental effectiveness of the therapies, that is more efficient than a parallel design in terms of required sample sizes. The objective is to test whether a subsequent therapy adds any significant benefit in treating the condition. Subjects will continue onto next therapy in the series if they failed a previous stage therapy. Conditional testing of hypothesis can be made sequentially that makes full use of all tested subjects.
