Sequential monitoring with administrative, ethical and economic advantages has become a standard in clinical trials. Adaptive designs including response adaptive randomization, covariate adaptive randomization and covariate adjusted response adaptive randomization has attracted lots of attention in the literature. Adaptive designs are also often desirable in clinical trials due to its ability to achieve certain ethical and efficient objectives. As a result, of interest is how to combine these two sequential procedures, make use of advantages of both procedures, and avoid possible problems such as the inflation of type I error rate. In this talk, we discuss the fundamental theoretical properties of this sequential procedure, investigate the approaches to control type I error rate, and demonstrate the advantages through numerical studies.
