Abstract:

Median duration of progression free survival is often forms a crucial component in the comparison of two cancer regimens when trying to evaluate the benefit provided by a new treatment. Unfortunately, progression, the main component of this endpoint, is only assessed on a periodic basis which can lead to an unstable estimate of the difference in treatment group medians. We explore the value of adding progression evaluations or randomizing patients to non-overlapping schedules. We also explore the ability of parametric and nonparametric interval-censoring methods to improve estimation. A randomized scan schedule appears helpful in improving median comparisons.
