We focus on the application of optimal experimental design methods in pharmacokinetic and combined pharmacokinetic/pharmacodynamic studies. After introducing the underlying nonlinear mixed effects regression model, we outline the numerical procedure of optimal design construction which is reduced to the optimization problem in the space of information matrices of candidate sampling sequences. Various population design software packages, including PkStaMp library, are discussed. Several approximation options for the individual information matrix are described, with special attention given to Monte Carlo-based option.
