Abstract:

A common way to achieve monoclonal cell lines is via limiting dilution and repeated cycles of subcloning. However, there is no standardized procedure that can easily tell the scientists how many subcloning cycles are needed to get monoclonal cell lines. Probability of single clone is often used as the criterion. This probability gets higher and higher with more subcloning cycles. The subcloning can be stopped at the cycle with high enough probability of single clone. Considering the inevitable variations during the limiting dilution process, I propose to use the simplified formula to calculate the probabilities of single clone at each clone cycle. An example is given for demonstration purpose.
