As a potential first step in the safety assessment of nongenotoxic carcinogens, the C-Path Carcinogenicity Working Group (CWG) designed and implemented a quantitative PCR-based signature to predict nongenotoxic compounds that induce liver tumors in the rat (Fielden et al. 2011 Toxicol. Sci.). Although this signature is specific to the application of predicting toxicity, we developed a computational workflow that is relevant to model building in different biological contexts. This presentation will discuss the challenges typical in biomarker studies within the context of predicting nongenotoxic hepatocarcinogens: building an unbiased predictive model; evaluating model precision; and establishing model equivalency between platforms.
