Abstract:

In this paper, genome-wide association study is applied in order to identify the genetic regions that are associated with the acute graft- versus-host disease (aGVHD). First, an empirical Bayes model is used to fit the pooled genotype data obtained by the DNA pooling technique. Then a Z-test is constructed to detect the SNPs that have significantly different minor allele frequency (MAF) between the case and the control group. To reduce the effect of the DNA pooling bias, the validation test is constructed to exclude the SNPs which have large bias. The haplotype information is also used to increase the power of the association test. A list of candidate genetic regions has been created so that the candidates can be confirmed by the further study of individual genotyping.
