Abstract:

Latent population substructure (LPS) is common in human genetic studies. It is well known that ignoring the LPS population based case-control genetic studies can lead to both an abundance of false discoveries in the form of spurious association and a serious decrease of power and resulting in false negative results. The loss of power is particularly serious when a low alpha-level is used as in typical genome-wide association studies (GWAS). To account for LPS in genetic association studies, we propose an application of the binomial mixture models and its associated likelihood ratio test in detecting disease-SNP associations in case-control association studies including GWAS. Numerical studies are used to illustrate the performance of the proposed models and methods.
