Abstract:

Studies of drug combinations have become increasingly important, especially in treating malignant cancers. Researchers are interested in identifying compounds that act synergistically when combined. The synergistic effects are generally evaluated by Loewe additivity model or by Bliss independence model. Although several review papers have discussed the scenarios where these methods should be applied, however, none of the discussions was originated from a statistical point of view. It still remains unclear to statisticians which model is more appropriate to use for a given data set. In this presentation, we will use numerical simulations and real data sets to compare these two models. A general guideline on how to properly conduct drug combination analysis will be given.
