Abstract:

In anticipation of the availability of next-generation sequencing data, there has been increasing interest in association analysis of rare variants (RVs). Due to the extremely low frequency of a RV, it is generally desirable to combine information across multiple RVs. Depending on the unknown distribution of the true association and non-association effects across multiple RVs, e.g, whether there are opposite association directions and how many non-associated RVs are present, various tests can be powerful, but no one is expected to perform uniformly well. Hence it would be desirable to develop versatile and adaptive tests that can maintain high power across various situations. We present some new data-adaptive tests and show their competitive performance as compared to existing tests.
