Abstract:

Combination drug products for multiple-target therapies are an important strategy for the pharmaceutical industry. However, such biological systems can have complex dose-response surfaces. Furthermore, some novel compounds may be weak when used alone, but substantially synergistic when used with another compound. Such complexity can cause difficulties for attempts to fit classical synergy models. However, nonlinear blending provides a general approach to combination drug synergy assessment that is applicable to any type of dose response surface. This talk will review the concept of nonlinear blending and discuss some practical aspects of its implementation for synergy assessment.
