Over the past few years association analysis has become the primary study design for finding new disease genes. Recent progress in sequencing technologies provides an unprecedented detailed view at the underlying genetic variation in humans, allowing for the first time a comprehensive assessment of the influence of the genetic variation on disease. This talk will review some of the challenges in the analysis of these data, and will focus in particular on recent developments for family-based association methods for large-scale genetic sequencing studies.
