Abstract:

Several multiplicity adjustments with strong control or weak control of the familywise error rate (FWER) were reviewed and illustrated for typical scenarios where multiple treatments (doses) and multiple endpoints are employed in a clinical trial. Strategies were considered with both strong and weak control of the FWER. These included the serial gatekeeping procedure, the gatekeeping procedure based on the Dunnett test, and the truncated Hochberg multiple test procedure (MTP) for strong control, and parallel step-down testing procedure and Hochberg testing procedure, are also considered for weak control. Property of each procedure is evaluated using simulations. Some discussions on choosing multiplicity strategy are provided for the practical implementation.
