Abstract:

We discuss here statistical models and methods for addressing some of the challenges that arise with the new data types, including those from second-generation sequencing. We will present a framework for the analysis of sequence data where we harness population genetics theory to provide prior information on effect sizes that allows a general and powerful test for association. The motivation for the proposed model is that underlying a belief in selectively rare causal SNPs is a correlation between the effect of a SNP on phenotype and its effect on the reproductive fitness of carriers. We contrast our approach with others where assumptions about the underlying genetic model are implicit.
