Abstract:

Increased worldwide collaboration amongst clinical trialists is leading to more opportunities for meta-analyses. To this point in time, standard clinical trial assumptions of proportional hazards with use of log-rank tests, and Cox regressions have been carried over into the meta-analysis setting. There is mounting evidence that such an assumption may lead to misspecification of model, the magnitude of effect on estimation in such a situation is largely unknown. ACCENT, the world's largest adjuvant colon cancer database, was used in investigations of a parametric (log-normal) framework and efficacy estimates were compared to those obtained with Cox model and Kaplan-Meier.
