Abstract:

The FDA Guidance on Antiviral Product Development recommends that sponsors of new antiviral agents "evaluate the in vitro antiviral activity of investigational products in two-drug combinations with other products approved for the same indication." A popular method among virologists to assess interactions between drugs is the isobologram. To make this analysis accessible to our scientific colleagues, we implemented it in MS Excel using the curve fitting add-in XLFit. We outline our estimation of the isobologram and illustrate using combinations with a NNRTI (anti-HIV agent). We highlight the need for data visualization and quality control of curve fitting. We compare the isobologram analysis with an alternative analysis, MacSynergy. We conclude by discussing strengths and weaknesses of the isobologram method, and identify opportunities for future method development.
