Abstract:

Many diseases, including cancer, are driven in whole or part by DNA copy number aberrations. The identification of aberrant regions is important to aid the medical and therapeutic research. A Bayesian model called Bayesian Segmentation Method (BSM) is proposed. The model jointly estimates locations of breakpoints, mean log2 intensity ratio and standard deviation of each segment. Significance of each segment is derived based on posterior distributions. Methods for selecting number of segments are demonstrated and compared. Using simulation data in Lai et al. (2005), BSM is compared with a number of methods including those concluded to be good in Lai et al. (2005) and a couple of new methods proposed after the paper. BSM is shown to be one of the best methods under different aspects. BSM is then applied to Glioblastoma Multiforme (GBM) from Bredel et al. (2005) for demonstration.
