Abstract:

Public microarray datasets have aided the increase in integrating multiple experiments via meta-analysis. In majority of these studies the new analysis examines the same question as the original studies. We examine statistical issues when the main experimental aim is to integrate datasets with a different experimental foci, i.e. to consider different questions. In particular combining different platforms emphasizing gene selection stability, a problem in most microarray analysis but intensified in integrative analysis. We propose a new stability score based on cross validation to identify stable and robust differentially expressed genes in datasets originally developed for a separate purpose. Our method was applied to research into the polygenic condition, hypertension where the main question studied by the scientific investigator was not previously addressed in the original experiments.
