Abstract:

The effect of CYP3A5 and other gene polymorphisms on the pharmacokinetics (PK) of the antiretroviral drugs, atazanavir and ritonavir, was determined using non-linear mixed effect modeling. Thirty-one HIV-negative adult volunteers participated in a 14-day longitudinal study, conducted at the University of Colorado Denver, General Clinical Research Center, to investigate the PK of these drugs. The study design was balanced by sex, race and CYP3A5 expressor status. Polymorphisms for other relevant drug transporter and metabolism genes were also measured. The Population PK analyses estimate typical values of parameters in the population, their variability, and factors such as genetic polymorphisms that explain this variability. Analyses utilize NONMEM software. Results will be compared to those obtained using the traditional two-stage PK summary approach.
