Abstract:

EWOC is a Bayesian method for selecting dose levels in Cancer Phase I Clinical Trials while controlling the probability of exceeding the maximum tolerated dose. Earlier implementation of EWOC is available as stand-alone Windows software, programmed in FORTRAN. The present implementation uses a novel "cloud computing" environment, and can be accessed as a web application, running R code on Apache web server. Using an adaptive dose escalation scheme, based on a parametric quantal response model, EWOC maximize the tolerated dose as fast as possible, subject to constraint that the predicted probability of assigning doses higher than the maximum tolerated dose, is equal to a specified value. Rapache is a module for the Apache web server, which allows embedding of R scripts in a webpage, connecting the HTTPD to R. MCMC methods are run through the BUGS project, accessed and controlled via R.
