Abstract:

This talk presents methods for evaluating an immunological biomarker(s) as a surrogate of protection in a placebo-controlled vaccine efficacy trial. The methods evaluate the biomarker as a potential surrogate endpoint for a right-censored primary endpoint. The methods require either baseline predictors of the immunological biomarker in a standard placebo controlled study, or, alternatively, a one-way crossover design wherein some placebo subjects are vaccinated and have their immunological biomarker measured. The methods can be used to compare biomarkers or to evaluate the value of additional biomarkers to improve surrogacy.
