Abstract:

This two-arm, phase II, proof of concept trial for a new anti-cancer compound, IL-18, in combination with Rituxan employs a Bayesian design to compare the complete response (CR) rate for IL-18+Rituxan to that of Rituxan alone. A non-informative prior is used for the distribution of the CR rate for the combination treatment. For Rituxan, a drug already on the market, an informative prior for the CR rate allows for a reduction in sample size for the control arm while maintaining overall power for the primary comparison between the two treatments. Simulation results demonstrating the properties of the design will be shown.
