Abstract:

Analysis of genome-wide association studies (GWAS) often beigns by examining simple main effects of the SNPs, one at a time. Although this approach has already led to the discovery of many susceptibility loci accross a variety of triats, it is evident that a better understanding of the full genetic architecture of the traits will require various types of additional analysis of GWAS studies. Examples include fine mapping of associated regions, studying associations for biological pathways and examintation of gene-gene and gene-environment interactions. The talk will review some of the scientific oppoprtunities and statistical challenges for such "value added" analysis of GWAS studies. Applications will be illustrated through various GWAS studies of cancers.
