Abstract:

We propose an extension of Bayesian logistic joinpoint regression model to design of Phase I clinical trials. We discuss the calculation of the unknown join points in the model corresponding to the individualized test doses. A numerical simulation shows the efficacy of the new approach in comparison with the traditional 3+3 design, CRM and other Bayesian models. The method is illustrated in a dose finding trial.
