Abstract:
|
In genetic studies, repeated measures or multivariate phenotypes related to a disease may be collected. In addition, family members are commonly recruited to examine the inheritance patterns, which results in multilevel clustering in the observations. To test the association between the genotype of a DNA variant and multivariate phenotypes while accounting for related individuals, approaches can be classified into: 1) regression models that allow correlated observations within a unit, 2) variable reduction methods such as principal components analyses and multivariate analysis of variance, and 3) combining univariate test results. We compare methods within and across the three approaches using simulation studies, and application of these methods to genome-wide association analyses is also discussed.
