Keywords: simulation ; optimisation ; clinical trial logistics
Abstract:

There are many sources of uncertainty in estimating the drug supply required for a trial. The modern trend to adaptive trials has added even greater uncertainties whilst also sometimes demanding more complex supply such as providing for a greater number of treatment arms. The problems of supply will be introduced and illustrated with examples from real trials. Then a structure for simulating and thus estimating supply needs will be described along with the difficulties of implementation, how it can be used, and future issues that need to be tackled. Finally an example of simulation for an actual trial will be presented.
