Abstract:

Many microarray experiments are designed to identify which RNA sequences are "differentially expressed" as compared to other sequences or between samples. In this study it is shown that, given enough within-array repetitions, most RNA sequences are significantly differently expressed, including randomly generated ones. It is therefore argued that it is more appropriate to search for sequences with expression levels which are significantly different from random ones. Regions of the genome, that are expressed in the target tissue, can be identified through tiled microarray experiments using this approach. The analysis of within-array replicates in this study indicates that within-array variability can be considerable and that this variability can be reduced with replication of probes within array.
