Abstract:
|
A drug exhibits linear pharmacokinetics (PK) when AUC(0-tau) from multiple dosing at steady state is equal to AUC(0-infinity) from single dosing (tau is the dosing interval length). The equivalence of these 2 AUCs can be tested using the confidence interval approach on transformed values. If this condition holds, the accumulation ratio, AUC(0-tau) from multiple dosing over AUC(0-tau) from single dosing, is equal to 1/(1-exp(-kel.tau)), where kel is the elimination rate constant. Individual effective half-life as ln(2)/kel can be derived from this method of calculating kel. Individual kel can also be computed from the terminal portion of the log linear plasma concentration versus time curve. Apparent terminal half-life can then be determined from this kel value. The statistical tools for assessing the PK to better estimate kel and half-life by these 2 methods will be discussed.
