Abstract:

In planning a multicenter trial, the choice between central and center-based randomization is often the tough one. Will center-based randomization provide a good balance in treatment assignments when a large number of permuted blocks on the randomization schedule are left incomplete? Can center-based randomization accommodate other stratification factors? Will stratification surely aggravate the incomplete block problem or could this be helped? Will central randomization require large drug stocks at the sites? Would a small percentage of forced allocations be acceptable? Are there ways of keeping the balance in treatment assignments---overall as well as within each stratum---while limiting the drug supplies at the sites? All these questions as well as possible solutions that combine elements of central and center-based randomization will be discussed at the table.
