Abstract:

The mean test for sib-pair allele IBD sharing is a widely used non-parametric statistical method to test for linkage in genetic analysis of family data. In 2002, I proved that the allele IBD sharing between sib-pairs are pair-wise independent, and derive a new method based on this result to calculate the non-parametric sib-pair mean test statistic. Compared to the traditional mean test, this new method calculates the variance of the statistic directly and does not rely on estimated variance of test statistics. The test statistic has a distribution with normal approximation, while the traditional mean test uses the observed values to estimate the variance of the allele IBD sharing, and therefore the test statistic has a Student's t-distribution.
