Abstract:

The Asthma Clinical Research Network (ACRN) was initiated by the NHLBI in 1993. When the ACRN exhausts its funding in 2010, it will have completed 15 randomized clinical trials. Two of the trials, BARGE and LARGE, recruited asthma patients with either the Arg/Arg genotype or the Gly/Gly genotype at the 16th position of the beta-agonist receptor gene. Both studies invoked a matched crossover design, and the properties of such a design are described. The research hypotheses for both studies were that patients with the Arg/Arg genotype would not respond to the administered beta-agonist medications as well as the patients with the Gly/Gly genotype. Finally, the primary results of the ACRN BARGE and LARGE trials are presented.
