Abstract:

The Women's Health Initiative postmenopausal hormone therapy trials demonstrated effects of estrogen plus progestin and estrogen alone on important clinical outcomes, and substantially changed medical practice. Here a high-dimensional SNP platform will be considered to identify subsets of women with relatively favorable benefit versus risk profile, and high-dimensional proteomic platform will be considered to identify biological pathways that help to explain clinical trial intervention effects. Methodologic issues related to platform throughput, multiple testing, and the approach to assessing biomarker changes as mediators of intervention effects, will be briefly discussed.
