Abstract:

Numerous genome-wide association studies (GWAS), where a large number of genetic markers are screened for association with phenotypes of interest, have been conducted and published in the last few years. Examples include studies of complex diseases such as age-related macular degeneration, atrial fibrillation, coronary artery disease, diabetes mellitus, amyotropic lateral sclerosis, multiple sclerosis, restless leg syndrome, and various cancers. How to best design and analyze GWAS is unclear and is the focus of intense current research. The design analysis strategies of recently published GWAS will be reviewed, discussing some of the proposed analysis approaches that attempt to bypass the limitations of the standard per-SNP analysis strategies. A look at what is ahead for GWAS and its role in integrated "omics" for understanding complex diseases will be included.
