Abstract:

The Cancer Genetic Markers of Susceptibility (CGEMS) initiative of the National Cancer Institute uses approximately 1100 cases and 1100 controls for each tumor type nested in prospective cohorts. Three stages relating to cohorts and statistical analysis are used in the strategy for identifying chromosomal regions. The first stage of the CGEMS project has been completed for both tumor types, providing genotype counts and p-vales under various statistical models for over 500,000 SNPs. In order to investigate over 40,000 DNA's, collaboration among multiple European and American groups were established. Over 10 susceptibility loci have been convincingly associated with prostate cancer. The risk-allele frequency may reach .85, showing lack of natural counterselection. Per allele odds ratios are small. Predicting cancer risk for patient management requires validation by large studies.
