Abstract:

Motivated by using stochastic differential equations for PK/PD model development, a method for developing PK/PD models based on the spline-enhanced differential equations is proposed. The new method allows information about the appropriate model structure to be extracted directly from data, which is accomplished by inclusion of flexible nonparametric functions in the ODEs to quantify model uncertainty. Furthermore, the new method provides the basis for systematic model development by allowing time-variations in key parameters to be tracked and visualized graphically. The performance of the new method is demonstrated by means of two examples with simulated data. Compared to SDE-based method, advantages of the proposed method are: intensive computation is avoided by using penalized least squares and the model misspecification can be modified along with the improved parameter estimates.
