Abstract:

A series of statistical models in functional mapping framework have been proposed to characterize quantitative trait loci (QTL) on the host genome that affect short-term HIV dynamics with a parametric function characterizing earlier stage of viral load trajectories. However, no particular mathematical functions have been available to describe long-term HIV dynamic changes. Here we present a novel statistical model for functional mapping that analyzes the whole HIV load trajectories semi-parametrically. The new model allows for the test of differences in the genetic control of short- and long-term HIV dynamics, and detects the effect of the viral-host genome interaction. We perform extensive simulation studies to test the statistical behavior of the model. The new model will provide an important tool for genetic and genomic studies of human complex diseases and their pathological progre
