Abstract:

We develop a score test for the relationship between longitudinal gene expression and ordinal event time data. The test utilizes longitudinal genearray histories to predict expression values at the time of the event. Longitudinally collected gene expressions are modeled by random effects whose distribution is left unspecified and conditioned out using the appropriate sufficient statistic (Tsiatis and Davidian 2001). We illustrate our method on a genomic dataset from a multicenter research project of inflammation and the host response to injury. This study aims to uncover the biological reasons why patients can have dramatically different outcomes after suffering a traumatic injury. The ordered response of interest is one of three states: death, no-change, or recovery of respiratory function. The longitudinal covariate is a genearray collected on seven scheduled times during one month followup. Our goal is to select a set of genes to be investigated further as potential longitudinal predictors of clinical events in patients who suffered severe traumatic injury.
