Title: Haplotype-based Association Studies on Multivariate Responses

Abstract:

Association studies are designed to identify markers that are tightly linked to genetic risk factors for complex diseases, such as asthma or diabetes. Many common diseases have complex phenotypes, and usually there are several clinical measurements analyzed simultaneously (e.g., glucose intolerance, dyslipidemia, high blood pressure). We propose a haplotype-based approach for quantitative multivariate phenotypes. We use a pseudo-likelihood function built using weights based on Gaussian kernels to estimate haplotype frequencies at every point in the response space. We discuss a novel multilocus measure of linkage disequilibrium and introduce tests for genetic association based on correlations between the multipoint linkage disequilibrium patterns and principal components in the response space. Finally, we discuss the accuracy and efficiency of the procedure.
