Title: Detecting Epistasis among the Quantitative Trait Loci

Abstract:

Epistasis, the interaction among genes at different loci, is an important factor that influences phenotypic expression of genes and genetic variation in populations. To study the presence of epistasis and its role in evolutionary biology, it is necessary to develop an effective and robust method to detect epistasis. In this paper, our focus is on statistical epistasis between two quantitative trait loci. We treat epistasis as the interaction in the two-way ANOVA model with three levels for both factors, and we propose a new method to detect it. We represent a model without the linear constraint on the interaction terms, typically used in two-way fixed effect models, and propose a test statistic to detect a non-zero interaction. In an identifiable situation defined in Terbeck and Davies (1988), our simulations show the new test has higher power than the classical F-test for interaction. We also present simulation results for general situations with an example.
