Title: Joint Localization of Two Linked Disease Susceptibility Genes Using Affected Relative Pairs

Abstract:

For complex diseases, there are often more than one disease susceptibility gene locating in a chromosome region. We develop a method that can simultaneously estimate the positions of disease loci and the genetic effects using affected relative pairs. The expected genes IBD at the marker locus is a function of the distance between the marker and the susceptibility gene and the genetic effects. The method applies generalized estimating equations (GEE), which is robust to model misspecification. As the asymptotic distribution depends on the number of independent pedigrees, which is much less than the number of affected relative pairs, we apply the small-sample adjustments in using the sandwich variance estimator in generalized estimating equations proposed by Pan and Wall (2002).
