Title: Statistical Methods for Inferring Epigenetic Phenomena Using Gene-expression Data

Abstract:

In cancer, events such as DNA methylation and histone modification play an important role in tumorigenesis. These are known as epigenetic events because they do not involve direct modification of the DNA sequence itself. While we commonly measure transcript mRNA as being on a high-throughput basis using microarray in cancer studies, how to infer epigenetic events from such data remains an elusive task. In this talk, we discuss progress in this area using cancer studies in which both measurements on copy number alterations and gene expression have been made. We study the issue of associations between copy number alterations and gene expression as well as inferring clusters of overexpression in microarray data and their ability to predict chromosomal amplification events. Examples from several cancer studies will be used to illustrate the proposed methodologies.
