Title: Statistical Design and Analysis in Systems Biology

Abstract:

Systems biology aims to study complex biological systems through high-throughput data. Identifying drug and/or disease biomarkers will result in a better understanding of drug efficacy and the predictive nature of animal models. We have multiple platform data collected on blood chemistry, transcripts, metabolites, and lipids from both preclinical and clinical samples. Experimental design issues such as sample size and power evaluation for multiple platforms and adjustment of confounding factors specific to omics platforms will be discussed. We have various challenges in the analysis: data preprocessing methods from various omics platform, the number of measurements much larger than the number of subjects, wide differences in the number of measurements per platform leading to imbalance in the platform contributions to the analysis, the issues of multi colinearity and multiple testing, and difficulty in proper model validation to avoid selection bias. We present statistical solutions and results on integration of various platforms to demonstrate the challenges and strengths of integrative analysis and the systems biology approach.
