Abstract:

Multiple analyses are common in clinical trials. Investigators often are asked to limit the number of primary analyses to a single question, even though this is often not practical in the clinical setting or helpful in evaluating related measures. The outcomes often are dependent on each other to varying degrees and differ in severity. The situation where the dependency is high (hyperdependency) is of major interest. The purpose of this work is to demonstrate situations where hyperdependency should be considered and to demonstrate its effect on hypothesis testing, sample size, and power analysis issues. Examples from clinical trial settings will be used to demonstrate its usefulness and to present guidelines regarding strategies for proper use.
