Title: Comparison of Links between Quantitative Trait Loci and Gene Expression Data

Abstract:

We developed a method to verify the link between the quantitative trait locus (QTL) mapping the drug abuse trait in mice and the set of genes associated with nucleus accumbens tissue identified by microarray. Biological evidence indicates the nucleus accumbens area of the brain is associated with the drug abuse trait. We compare the supposed link with a referential QTL set and a referential gene set not associated with the drug abuse trait. We introduce two perspectives to quantify the link between a set of QTLs and a set of genes. From each perspective, we primarily apply generalized linear model (GLM) and generalized linear mixed model (GLMM). Based on the selected models, we test the strength of the quantified link. Our analyses reveal the proposed link is not always significantly stronger than the referential links. We also do a simple simulation to verify the link. The results show the supposed link is stronger than expected by chance. However, the referential link also is significantly stronger than by chance. We hope the procedure provides a statistical methodology relevant to similar studies in the field of bioinformatics.
